John D. Barrow nasce a Londra nel 1952. Si laurea in Matematica presso la Durham University nel 1974 e consegue il dottorato in Astrofisica alla Oxford University nel 1977. Lavora presso la Oxford University e presso la California University di Berkeley prima di ottenere, nel 1981, un posto all'Astronomy Centre dell'Università del Sussex, dove, fino al 1999, è professore di astronomia e Direttore del Centro di Astronomia.
Autore di 300 pubblicazioni in materia di cosmologia e astrofisica, ha ottenuto riconoscimenti come il Locker Prize for Astronomy, la Kelvin Medal of the Royal Glasgow Philosophical Society nel 1999 e, nello stesso anno, una laurea honoris causa presso l'University of Hertfordshire. Recentemente ha ottenuto un assegno di ricerca da parte del Particle Physics and Astronomy Research Council del Regno Unito. Nel luglio 1999, è stato nominato Research Professor di Scienze matematiche a Cambridge e Direttore del Millenium Mathematics Project, una nuova iniziativa per diffondere l'apprendimento e far riscoprire il valore della matematica e delle sue applicazioni tra i giovani e nella società.
È autore di 12 libri, tradotti in 29 lingue, in cui esplora molti aspetti delle principali implicazioni storiche, filosofiche e culturali dell'astronomia, della fisica e della matematica: The Left Hand of Creation, scritto con Joseph Silk (La mano sinistra della creazione, Mondadori 1985), L'Homme et le Cosmos (1984, scritto con Frank Tipler), The Anthropic Cosmological Principle (1986, scritto sempre con Tipler), The World Within the World (Il mondo dentro il mondo, Adelphi 1991), Theories of Everything (Teorie del tutto, Adelphi 1992), Pi in the Sky: counting, thinking and being (La luna nel pozzo cosmico, Adelphi 1994), Perché il mondo è matematico? (Laterza 1992), The Origin of the Universe (Le origini dell'universo, Rizzoli 2001), The Artful Universe (L'universo come opera d'arte, Rizzoli 1997), Impossibility: the limits of science and the science of limits (Impossibilità, Rizzoli 1998), Between Inner Space and Outer Space (Dall'io al cosmo: scienza, arte, filosofia, Raffaello Cortina 2000), The Book of Nothing (Da zero a infinito, la grande storia del nulla, Mondadori, 2002). Un nuovo volume, The Constants of Nature, è in corso di pubblicazione.
Conferenziere di fama internazionale, ha partecipato, non solo nel Regno Unito, a importanti convegni scientifici come le Gifford Lectures (1989), la George Darwin Lecture of the Royal Astronomical Society, l'Amnesty International Lecture on Science a Oxford, The Flamsteed Lecture, The Tyndall Lecture, The RSA Christmas Lecture for Children e la Spinoza Lecture presso l'Università di Amsterdam. John Barrow ha anche il singolare merito di aver tenuto incontri sul tema della cosmologia al Festival del Cinema di Venezia, presso la residenza del primo ministro inglese in Downing Street, al Castello di Windsor e in Vaticano.
